I new to the whole RV thing myself....but....
I try to make sure to run the A/C, top up the batteries and let the motor warm up once a month if I haven't made any trips.

I never let mine sit more than 30 days, that keeps it running good. Winter is Glamis, and summer is Needles with the boat.

check everything once a month keep enuff gas to run generator for a few minutes keep batteries charged i would recomend either disconecting them or making sure all parasitic draws are unplugged. like dvd players or anything in cig lighters. ( they drain the batteries when left on for a month. also run water thru pump every so often. for use check tires always check tires every day you drive inspect pressures check tread every morning of driving( trust me). check windows they seal and dont leak rain water. hose water is acceptable. lube every hinge with silicone lube. they will only squeak in the middle of the night. check for drafts under the mattresses. check drive shaft u joints for wear.check shocks, they rust from sitting idle.this is some of the stuff I found works, good luck.

How many miles on this?
How old is it?
Make sure if you don't have written log of when things were changed, change the ATF right away.
The TH400 is a lot stronger than the 4L80e (the overdrive transmission), but in either case it should be changed.
ALSO: add a transmission temperature gauge NOW before you tow.
With that, you will avoid VERY costly transmission replacements.

I have a gasser (V10). I run synthetic oil mainly because of the weight. I like to think it lubricates a little better under loads and also it might save a little gas since it isn't as heavy as conventional oil. I try to start the generator and motor at least once per month but usually I dont need to think since I pull the boat with it to the river in the summer and the bikes in the winter.

454 is a strong engine. Run it when you can, keep the batteries charged. change all the fluids to be safe and I agree with the previuos post as to switch to Synthetic. as far as the rig, start up everthing before each trip. Drain the hot water heater and change the Anode rod.
